CHARACTERIZATION OF THE UTERINE ENVIRONMENT IN ACYCLIC EMBRYO RECIPIENT MARES

Nineteen healthy mares in seasonal anestrus were evaluated when subjected to long-acting progesterone at two different doses. Based on these conditions, two experimental groups were formed and evaluated at different times. The following characteristics were studied: plasma P4 concentration by radioimmunoassay (RIA), protein concentration present in the uterine fluid obtained by low-volume uterine lavage using the bicinchoninic acid (BCA) method, as well as protein profile by one-dimensional electrophoresis (SDS-PAGE 12%). The bands of interest from the electrophoresis were subjected to trypsin digestion and analyzed by mass spectrometry (LC-MSE) for subsequent identification by the BLAST/NCBI tool. The results demonstrated that the application of estradiol benzoate increased the plasma progesterone concentration from 0.55 to 30.15 ng/mL in G1 (p<0.05), and from 12.03 ng/mL to 16.50 in G2, with no significant difference. There was no significant difference in the plasma progesterone concentration between the days evaluated in G2. In G1, no significant difference (p<0.05) was observed for the protein concentrations in the uterine fluid. In G2, the mean protein concentration on D4 (0.31µg/µL) was lower when compared to the mean on D9 (0.78µg/µL). Electrophoresis and spectrometry demonstrated the appearance of uterocalin in both groups on D4, up to D15 and D9 of G1 and G2, respectively. The presence of Lipocalin 2 and uteroferrin was observed only on D15 of G1. The expression and concentration of uterine proteins were dependent on the administered dose of P4LA in anestrous mares.
