The aim of this study is to analyze the role of nursing in the guidance and monitoring of newborns with Glucose-6-Phosphate Dehydrogenase (G6PD) deficiency, highlighting educational interventions and comprehensive care strategies. The methodology used is an integrative literature review. Studies published between 2019 and 2025 in Portuguese and English were included, totaling 15 publications, including scientific articles and official protocols. The results demonstrated that nursing plays a central role from neonatal screening, through the heel prick test, to continuous outpatient follow-up. Guidance on factors that trigger hemolytic crises and health education stood out as essential actions for guiding caregivers, preventing complications, and promoting the child's quality of life. It is concluded that evidence-based nursing care is based on qualified listening, which is essential for the safe management of G6PD deficiency.
